SYP Antibody (D-4) is an IgG1 κ mouse monoclonal SYP antibody (also designated SYP antibody, SYPH antibody, Major Synaptic Vesicle Protein P38 antibody, MRX96 antibody, or MRXSYP antibody) that detects the SYP protein of mouse, rat and human origin by WB, IP, IF, IHC(P) and ELISA. SYP Antibody (D-4) is available as both the non-conjugated anti-SYP antibody form, as well as multiple conjugated forms of anti-SYP antibody, including agarose, HRP, PE, FITC and multiple Alexa Fluor® conjugates. Synaptic vesicles participate in a cycle of fusion with the plasma membrane and reformation by endocytosis. Synaptic vesicle protein synaptophysin (SYP) is targeted to early endosomes in transfected fibroblasts and in neuroendocrine cells. SYP is an N-glycosylated intergral membrane protein found in neurons and endocrine cells that associates into hexamers to form a large conductance channel. SYP contains four transmembrane domains and may function as a gap juction-like channel. Membrane cholesterol specfically interacts with SYP to play a role in vesicle formation. Synaptobrevin (VAMP) also binds to SYP and the resultant complex is upregulated during neuronal development, but is absent in exocytosis fusion complex. Thus, the synaptophysin-synaptobrevin complex is not essential for exocytosis, but rather provides a pool of synaptobrevin for exocytosis. In addition, the tail domain of brain Myosin V also forms a stable complex with synaptobrevin II and SYP, and this complex is disassembled upon the depolarization-induced entry of Ca2+ into intact nerve endings.

We used the SYP(D-4) with the mouse IgG binding protein (488) on fixed mouse brains but did not see any signal. What can you recommend? Do we need a specific antigen retrieval step for this antibody or should we try traditional secondary antibodies?

Can you recommend a positive control to use with SYP (D-4): sc-17750 monoclonal antibody for Western blot?

We recommend IMR-32 (sc-2409) or MDA-MB-231 (sc-2232) whole cell lysate, rat brain extract (sc-2392) or mouse brain extract (sc-2253). whole cell lysates to use as positive controls as we have Western blot data showing specific detection of SYP in these cell lines.

What secondary is best to use with SYP (D-4): sc-17750 monoclonal antibody?

It gives a strong and specific signal corresponding to the expected molecular weight in both murine and human differentiated neuroblastoma cell lines. Antibody dilution 1:1.000, overnight incubation at 4°C in TBST + 5% no-fat dry milk.

In altri modi non funziona in cellule che si differenziano verso il fenotipo neuroendocrino, non cellule neuronali. Il secondario, invece, era utilizzato alla concentrazione di 1:2000 in milk al 3%
