Chk1 Antibody (G-4) is an IgG2a κ mouse monoclonal Chk1 antibody (also designated CHEK1 antibody, or checkpoint kinase 1 antibody) that detects the Chek1 gene product of mouse, rat and human origin by WB, IP, IF, IHC(P), FCM and ELISA. Chk1 Antibody (G-4) is available as both the non-conjugated anti-Chk1 antibody form, as well as multiple conjugated forms of anti-Chk1 antibody, including agarose, PE, FITC and multiple Alexa Fluor® conjugates. Cell cycle events are regulated by the sequential activation and deactivation of cyclin dependent kinases (Cdks) and by proteolysis of cyclins. Chk1 and Chk2 are involved in these processes as regulators of Cdks. Chk1 and Chk2 both function as essential components in the G2 DNA damage checkpoint by phosphorylating Cdc25C in response to DNA damage. Phosphorylation inhibits Cdc25C activity, thereby blocking mitosis. Cdc25A, Cdc25B and Cdc25C protein tyrosine phosphatases function as mitotic activators by dephosphorylating Cdc2 p34 on regulatory tyrosine residues. It has also been shown that Chk1 can phosphorylate Wee1 in vitro, providing evidence that the hyperphosphorylated form of Wee1, seen in cells delayed by Chk1 overexpression, is due to phosphorylation by Chk1.

Has this antibody been tested for the sole kinase domain as well (approx. residues 1-270)?

Does this antibody recognize hamster total Chk1?

Thank you for your question. We do not have any data or product citations using this antibody, Chk1 (G-4): sc-8408, for detection of Chk1 in hamster. The antibody is raised against full-length Chk1 of human origin, and based on a predicted protein sequence for Chk1 in hamster (accession number: XP_003511081) this sequence shares 95% sequence identity with the hamster protein. That is a significant, and if the predicted sequence is correct, then there is a very good chance the antibody may also cross-react with hamster.
