CD63 Antibody (MX-49.129.5) is an IgG1 κ mouse monoclonal CD63 antibody that detects the CD63 protein of mouse, rat and human origin by WB, IP, IF, IHC(P), FCM and ELISA. CD63 Antibody (MX-49.129.5) is available as both the non-conjugated anti-CD63 antibody form, as well as multiple conjugated forms of anti-CD63 antibody, including agarose, HRP, PE, FITC and multiple Alexa Fluor® conjugates. The tetraspanins are integral membrane proteins expressed on cell surface and granular membranes of hematopoietic cells and are components of multi-molecular complexes with specific integrins. The tetraspanin CD63 (also known as LAMP-3, Melanoma-associated antigen ME491, TSPAN30, MLA1 and OMA81H) is a lysosomal membrane glycoprotein that translocates to the plasma membrane after platelet activation. CD63 is expressed on activated platelets, monocytes and macrophages, and is weakly expressed on granulocytes, T cell and B cells. It is located on the basophilic granule membranes and on the plasma membranes of lymphocytes and granulocytes. CD63 is a member of the TM4 superfamily of leukocyte glycoproteins that includes CD9, CD37 and CD53, which contain four transmembrane regions. CD63 may play a role in phagocytic and intracellular lysosome-phagosome fusion events. CD63 deficiency is associated with Hermansky-Pudlak syndrome.

A number of reviewers mentioned good western detection of CD63 from non-reducing exosomes. Does the antibody work only on non-reducing CD63? What would be the purpose of running non-reducing exosomes on SDS-PAGE? Thanks!

I am researching secreted exosomes, generated from intra-luminal vesicles present in endolysosomes. Would the epitope of this antibody be available to this antibody in this sample?

Can this antibody be used for flow cytometry? And if possible, what is the optimal concentration for this technique?

Thank you for your question. Anti-CD63 Antibody (MX-49.129.5) is recommended for detection of CD63 of mouse, rat and human origin by flow cytometry. We recommen to use 1 μg per 1 x 106 cells.

Hello, I would like to use this antibody to detect CD63 in fetal bovine serum. Is there any data available showing the corss reactivity with bovine CD63? Thank you!

Thank you for your question. BLAST analysis shows an 85% sequence homology between the full length sequence of human CD63 and bovine CD63, which is lower than our minimum percentage for species reactivity recommendation. Unfortunately, we do not characterize the exact binding site of this monoclonal antibody and we have not tested it with bovine samples so it is uncertain that the antibody will react with the bovine target. Does this antibody (CD63 Antibody (MX-49.129.5) recognize a cytosolic or luminal/extracellular domain of CD63? i.e has it been epitope mapped?
