Smad2/3 Antibody (C-8) is an IgG2a κ mouse monoclonal Smad2/3 antibody that detects Smad2 and Smad3 of mouse, rat and human origin by WB, IP, IF, IHC(P) and ELISA. Smad2/3 Antibody (C-8) is available as the non-conjugated anti-Smad2/3 antibody form, as well as multiple conjugated forms of anti-Smad2/3 antibody, including agarose, HRP, PE, FITC and multiple Alexa Fluor® conjugates. Smad proteins, the mammalian homologs of the Drosophila mothers against decapentaplegic (Mad), have been implicated as downstream effectors of TGFβ/BMP signaling. Smad1 (also designated Madr1 or JV4-1) and Smad5 are effectors of BMP-2 and BMP-4 function, while Smad2 (also designated Madr2 or JV18-1) and Smad3 are involved in TGFβ and Activin-mediated growth modulation. Smad4 (also designated DPC4) has been shown to mediate all of the above activities through interaction with various Smad family members. Smad6 and Smad7 regulate the response to Activin/TGFβ signaling by interfering with TGFβ-mediated phosphorylation of other Smad proteins.

I want to detect SMAD2/3 phosphorylated protein expression in porcine cells. But I'm not found this criteria in primary antibody of Santa cruz. I found that p-Smad2/ 3 (Ser 423/425) is not available now. Can you suggest me which one is proper?

I WOULD LIKE TO PURCHASE ANTIBODY AGAINST PHOSPHORLYATED SMAD1 AN SMAD2 AND THEIR SECONDARY ANTIBODY FOR WESTERN BLOT

Thank you for your question. Unfortunately, we do not currently offer any phospho-specific Smad1 or Smad2 antibodies. This mouse monoclonal, Smad2/3 Antibody (C-8): sc-133098, is against total Smad2/3 and may detect both the phosphorylated and non-phosphorylated forms of the proteins.

Can Smad2/3 (C-8): sc-133098 monoclonal antibody be used for detection of Smad2/3 of equine origin?

Rated 5 out of 5 by IuriCV from Nice and strong band (w.b.) Produced a single band at the expected molecular weight (~55kDa). Sample: MDA-MB-231 wcl (30ug). Blocking: 1h in blocking buffer (0.05% tween-20 in TBS with 5% skin milk). Primary antibody: 1:200 in blocking buffer, overnight @ 4oC. Secondary antibody: 1:3000 in blocking buffer. Exposure time: 5 seconds* I'm certainly very satisfied with this product. *further dilution of the primary antibody, maybe even at 1:1000, will likely yield similar results at the expense of a prolonged (and more adequate) exposure time.

Rated 5 out of 5 by mingwunl from my review for Smad2/3 antibody (sc-133098) I tested a free antibody for Smad2/3 antibody (sc-133098) using IP samples from mouse fibrotic kidneys for Western blot. We loaded 80 ug proteins in each slot in the SDS gel. The antibody was diluted in 1:500 in 3% BSA/PBST and incubated overnight with the blot. We can observe a very clear band around 55-60 kd. We have repeated this experiment several times and used the fresh diluted antibody on the fresh blot.
