HLA class I Antibody (HP-1F7) is an IgG1 κ mouse monoclonal HLA class I antibody (also designated HLA major histocompatibility complex, class I antibody, major histocompatibility complex (MHC) class 1 antibody, chromosome 6p21 gene complex class 1 antibody, or 45 kDa transmembrane class I human leukocyte antigen antibody) that detects the HLA class I protein of human origin by WB, IP and FCM. HLA class I Antibody (HP-1F7) is available as both the non-conjugated anti-HLA class I antibody form, as well as multiple conjugated forms of anti-HLA class I antibody, including agarose, HRP, PE, FITC and multiple Alexa Fluor® conjugates. The major histocompatibility complex (MHC) is a high genomic density gene family that plays an important role in the immune system, autoimmunity and reproductive success. Human MHC genes are referred to as human leukocyte antigen (HLA) genes. MHC class I molecules consist of two polypeptide chains, an a or heavy chain, and β-2-Microglobulin, a non-covalently associated protein. Cytotoxic T lymphocytes bind antigenic peptides presented by MHC class I molecules. Antigens that bind to MHC class I molecules are typically 8-10 residues in length and are stabilized in a peptide binding groove. Accumulating in endosomal/lysosomal compartments and on the surface of B cells, HLA-DM and -DO molecules regulate binding of exogenous peptides to class II molecules (HLA-DR) by sustaining a conformation that favors peptide exchange. The differential structural properties of MHC class I and class II molecules account for their respective roles in activating different populations of T lymphocytes.

I have done computational analysis for HLA-A*01:01 interaction with peptide LSWEMELAY, now I want to validate it experimentally. Can I use HLA class I Antibody (HP-1F7): sc-69892 for my study?

Can HLA class I (HP-1F7): sc-69892 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, HLA class I (HP-1F7): sc-69892 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.
